MPO-ANCA (capture technique)

Indication

Suspicion of systemic vasculitis, especially with kidney involvement.

Method

Capture ELISA with granulocyte myeloperoxidase (MPO) as antigen.

Accredited analysis – read more at the Quality page.

Answer

The result is given as international units (IU/ml).

Interpretation

Capture MPO-ANCA is a complementary method in the analysis of MPO-ANCA, where a monoclonal antibody is used to catch MPO instead of having the protein bound directly to the plastic surface. The monoclonal antibody is chosen so that it blocks an epitope of anti-MPO antibodies, which gives C-ANCA that is not related to vasculitis. This makes the analysis more specific for vasculitis. The capture method has a higher sensitivity than direct ELISA and therefore can detect much higher concentrations in individual patients.

MPO-ANCA is a marker for the vasculitis diseases granulomatosis with polyangiit (GPA), previously named microscopic polyangiitis, and eosinophil granulomatosis with polyangiitis (EGPA), previously named Churg-Strauss syndrome. Antibodies level may decrease during remission and an increasing level may suggest recurrence. MPO-ANCA may also occur in SLE, infections and drug induced conditions without relation to vasculitis.
